RingCap® technology
RingCap® (Ring-Link Capture Library Technology) is a patented technology independently developed by SpaceGen. It pertains to a targeted sequencing library preparation technology based on multiplex PCR, which addresses many limitations in the preparation of targeted libraries using traditional PCR technology, such as complex experimental procedures, long manual operation time, and high upfront equipment investment.
Core Technology—RingCap® Technical Principle
The First Stage:
1. Modified specific primers and highly specific Spacegen - Taq enzymes were employed to amplify and capture the target sequence, resulting in the formation of a sticky end in the product.
2. DNA ligase was utilized to link the asymmetrically ligated probe to the sticky end, thereby forming a circular product to prevent binding between the products.
The Second Stage:
Specific primers were used to specifically amplify the circular product, enabling the acquisition of the target sequence and, simultaneously, the binding to the required specific marker sequence.
